I need to co-cultivate marine actinomycetes (3% NaCl requirement) and freshwater live Aeromonas for the induction of antimicrobials production. What broth could I use?
I've no great experience with actinomycetes, but any nutrient-rich broth, as BHI or Nutrient broth are OK for Aeromonas cultivation. I'd try first to culture the microorganisms separately to check if they are able to grow, cause 3 % NaCl can be inhibitory to aeromonads depending upon the strain and the temperature.
For actinomycetes, dry samples by heat treatment (remove non-spore formng bacteria) and spread diluted samples (with sterilized seawater) on agar media.
Screen freshwater Aeromonas capable of growing at 3% NaCl. It should be very easy to do and you will find a lot of such organisms. You may even take a standard strain from a reputable culture collection and do the screening. If you do not get one then adapt that standard culture to grow at 3% NaCl. Again very simple to do bu then do it in a step wise manner. In spite of this you may not succeed in co culturing the Aeromonas and actinomycete. This is due to the antibiotics the actinomycete might produce. Therefore, be careful.
