I am performing soil enzymatic activity assay using chemical methods.The enzymes mainly I targeted are DHA,Urease,Protease,Soil Phosphatase and B-Glucosidase.I performed the experiments but having problem in calculating the enzymatic activity.Supoose if we consider DHA, I got the abosrbance values of my sample and then extrapolated to standard curve of formazan(end product if substrate is TTC) to find out the unknown concentration.For eg. I got the concentration as 9 ug of formazan released/gram of soil in 24 hours.So is this my enzymatic activity or I need to calculate the activity using formula where dry weight,reaction volume will be taken in to account.Please help.
Same thing goes with other enzymes also.

Soil enzyme activity is an index to show that the microbial population is ample to convert the chemical to available forms to the plants. It is a combination of phosphatase activity, nitrogenase activity etc., which also tells about the soil health status on application of that particular pesticide.
you have calculated the TPF concentrations calculated from the calibration curve , the DHA= (TPF(concentration)*V ) / (dwt* w.)
V=the volume of solution added to the soil sample
dwt=is the dry weight of your sample( you have first to calculate the dry matter )
w=The moist soil used(g)
