How to calculate SOD enzyme activity per gram of seed?

I measured activity of SOD enzyme with NBT method by Spectrophotometer.

how i can transform the initial absorbance to Unit/g of seed

Please explain step by step method for learning this subject.

The information that I have obtained from Spectrophotometer are the following:

The absorbance of Control at 560 nm= 0.453

The absorbance of Sample at 560 nm = 0.220

Total volume buffer extraction= 6ml

Total reaction volume for read the absorption= 2.8mL (20ul enzyme extract used for reaction)

seed sample for extraction=1 gram
