i use according to attached method for lysozyme assay. but I don't know how to calculated enzyme activity and unit of enzyme.how should I use wavelengths that have developed and are evaluated lysozyme activity?
For the calculation of the lysozyme enzyme activity, you may use the following equation:
Units/ml enzyme = (ΔA450/min Test – ΔA450/min Blank) (df) / 0.001x Volume in mL of enzyme solution
where:
df = dilution factor
0.001 = ΔA450 as per the Unit Definition
To apply this equation to the study described in the file attached to your question, you should substitute ΔA450 with ΔA420 and Volume in mL of enzyme solution should be 0.5. Please note that the incubation time in the study described in the attached communication was 30 minutes. All of your calculations should be based on the results illustrated in Figure 2.

Hi. Thanks a lot Rafik. but what is it factor dilution in my assay? I don't use from any dilution in my sample! I pick supernatant (after centrifugation) and without making any dilution, apply according attached protocol.
I have dilution process in product standard curve of lycozyme with specific concentration. are this you're purpose about dilution factor or no?
