How to calculate Ksv and Ka in BSA fluorescence quenching?

Welcome everyone,

I am asking you to explain how to calculate in Origin Ksv and Ka of BSA fluorescence quenching? I have a nonlinear plot, so I cannot use the Stern-Volmer equation. I am attaching a photo of the plot.

I found that there is static and dynamic quenching and should be used the equation:

I0 / I = (1 + Kq x τ0 x [Q]) (1 + Ka x [Q])

The problem is that you apparently have both static and dynamic quenching and hence you cannot apply the simple Stern-Volmer equation. If you could measure lifetimes you could sort out these issues. Here is a chapter from my book Introduction to Fluorescence which discusses these issues.
