I am working on transforming suspended calli of Catharanthus roseusby Agrobacterium tumefaciens. According to the transformation efficiency formula (Transformation Efficiency (%) = (Number of transformed calli / Total number of calli) * 100), number of transformed and total callus is required. But in suspension culture of calli it is impossible for calculate the number of transformed calli and total calli. If anyone has experience in this field or can give me advice, I would be happy to use their experiences and advice.

To calculate transformation efficiency in suspended calli, use this formula:

Transformation Efficiency (%) =(Number of transformed calli / Total number of calli treated)× 100
For example, if you treated 100 calli and 25 were successfully transformed (e.g., show GFP signal or grow on selection media), then:
(25/100) × 100 = 25%
Just count how many calli were transformed and divide by the total you treated, then multiply by 100.
