When we develop a new method first we need to know the critical point during chemical reaction to avoid interference.

Them we use the sensor in standard well known analyte (in this case pure glucose from oficial company)

The last step is to compare and apply in real sample (serum in this case)

The answer is pretty simple. First, I believe you already did your calibration curves for glucose detection in simulated blood/serum or blood/serum spiked with known concentration of glucose (that can be done also using a standard addition method). Then, you can use your biosensor to measure glucose levels in real samples and correlate the levels that you measured to a standard method such as the glucometer or HPLC-MS. When you have these, you can build your Receiver Operating Characteristic curve that will tell you the characteristics of your biosensor.

I'd recommend using a Bland-Altman plot to compare your sensor to a standard detection technique. There are ELISA kits for the measurement of glucose or simply using a commercially available blood glucose monitor. Just find one that has a similar sensitivity limit and range of detection as your sensor. 

These alternative detection techniques serve as your system validation. Just make sure you are running these tests in parallel using the same samples on your biosensor.
