Can anyone suggest a practical way to identify and separate the primary root from clusters of lateral and adventitious roots in 1 month-old Arabidopsis plants pulled out of the soil?
I would suggest washing the roots to get the soil off and then floating them on a large petrdish full of water or liquid media. then you can lightly with a fine tip paint brush push the LRs away from the primary, and quickly take a picture
Thanks Rahul. But I was facing difficulty while doing the same. :-) LRs are so thickly clustered at times so that separating primary roots are getting really tough.
I think the better way is after washing up the roots you can mummies the roots because in this life stage the roots are so fine of course if you don't need the plants again! mummies the roots give you another chance to see and separate the fine roots.
