To check in vitro toxicity you can check the cell viability assay of your nanomaterials in different normal cells. And through in vivo also you can optimize the non toxic dose of your nanoparticles by checking the mortality, blood and serum parameters, histopathology, genotoxicity etc.

Biocompatibility is a relative concept, rather than absolute. It's important to consider the proposed application. Cytotoxicity is a good place to start, but is not enough to guarantee biocompatibility. Not killing the cells doesn't mean they'll behave the way you need them to.

A lactate dehydrogenase (LDH) assay (eg the kit available from Sigma) is a fairly simple and effective test for cell toxicity. If you don't have specifics in mind, and are new to cell culture, working initially with cancer-derived cells or dermal fibroblasts will be the easiest starting point.

Hi , you can better go with test with red blood cells if u have idea in taking this particle for application in the biological field...(i.e., for the application of using this particle for delivering or to bind or for targeting with the complexes for targeted delivery applications... for that u have to test the hemo biocompatibility of your particle for the particular time period of incubation with the red blood cells.(eg from minutes to hrs incubation)...if no morphological changes were observed in the shape of RBCs trough microscope, it is worth to report as biocompatible... or by spectroscopically measuring the OD at 415nm with UV spectroscopy....

When we test the biocompatibility of material especially for medical devices,

there are some categories depending on final usages.

For example, cytotoxicity, skin irritation, sensitization, systemic toxicity, and so on.

ISO 10993 describes the biocompatibility test for medical devices.

The requirements (test items) differ from the kind of medical devices. The categories are devided depending on "Contact duration time" and if the devices are "surface devices, externally communicating devices, or implant devices"

However, when you chek the biocompatibility of medical devices, I recommend to start with cytotoxicity. Most of cases, if the cytotoxicity is low, it is more probable to pass the rest of categories.

Your key words are "implant & metal", you may think of implantable metalic medical devices for permanant implant (more than 30 days) and implanted in Tissue or Bone.

In that case, ISO requires following biocompatibility test;

Cytotoxicity, Sensitization, Irritation, Acute systemic toxicity, subcronic toxicity, gentocicity, Implantation, Chronic toxicity, Carcinogenicity.

Another recent popular assay I would suggest you and that is CAM assay. You can check the in vivo biocompatibility assay in chicken egg. This is a very standard and easy experiment.

The thrombin generation action of your material is best studied by performance of a recalcified coagulation activity assay (RECA).

At initial level you can do in vitro cytotoxicity test for percentage cell viability. The type of cells used for evaluation depends on your application of material.
