Well, pharmacology is not really my field. I would suggest the usage of a good software. They may have commands to "translate" numerical data (generally given in tabular form) into a graphical representation.
Otherwise hand work has to be performed to have a formula for an approximate function.
Concentration-effect curves are usually plotted with a logarithmic x axis (concentration) and a linear y axis (effect), but in some cases both can be log. Although even Excel can be tortured into doing this, R is probably best, unless you have lots of money, in which case get commercial software.
GraphPad Software is useful to obtain dose-response curve, by ploting logarithmic x axis (concentration) and a linear y axis (% effect of control),

The most powerful tool is undoubtedly Wolfram_Mathematica if it is provided by your university / institution, but if you want a free SW you can try SIMFIT 7 downloadable at link below. See the attached introduction

You can use Sigma Plot software but we obtained excellent results by using GRAPH PAD software.
You do not need any software other than Excel to plot the graph and obtain the necessary equation. I have analyzed the dose response relationship for contractile effects of acetylcholine in isolated guinea pig ileum before and published the methodology in ResearchGate. Please see Section 8.1 of the attached file for detail.

In short :
For each concentration, produce the responces as means and s.e.m., express the concentrations on a p scale (as –log10M) on the abscissa (x axis), and the absolute values of the response (y axis) (and in another table and graph as the percents of maximum responses). You will use the percents to calculate EC50 and pA2. Anyway, you cannot learn it just from the books - or who knows, may be you can…

Actually i was doing MAO-B enzyme assay for different samples, first i used common inhibitors of MAO-B enzyme named selegiline. first i took different concentrations of selegiline according to smaller to higher concentration along with control and blank. from this dose response curve i took the value of EC50 for estimating the samples study. and i did the triplicate study of my in-vitro work. and i used the sigma plot software for measuring dose response curve.
