Different species of Heterodera need special conditions based on host specify but CCN hatch in 12C and in wet soils without host. I think you can conserve cyst in 4C but try to inoculate them as soon as.
You may hand pick the cysts under a steroscope from a soil sieved suspension, using a sharp tweezer or any other simple tool. Dry the cysts out on adsorbing paper and then store them dry in an eppendorf with some silica gel particle, for subsequent use for inoculation in pots or other container. If you have previously measured the mean number of viable eggs per cyst, you can add the required number of cysts in soil for the final nematode inoculation.
