I have isolated more than 300 bacteria from rhizosphere, roots epiphytes and endophytes. Now which test should i conduct on bacterial isolate which can help to improve drought tolerance of the plant.
By making some physiological changes in terms of the biological metabolism and secretion of certain hormones they will increase the tolerance of cells to stress especially the ROS group
A possible mechanism is lowering plant growth rate. The same that is used in drought resistant GMOs. When plant is growing slower it need less water and survives better in drought conditions.
Now for sure you have to test these 300 strains for drought resistance and select the most drought resistant ones, especially those that maintain their usefulness (for example N fixation, etc) under drought conditions. The next step is greenhouse, and then field investigations with the crop of your choice.

I suggest to perform ACC-deaminase activity of the isolated strains (either by biochemical method or by molecular anal). It is well established in literature that those strains which have ACC-deaminase activits, can help plants to mitigate stress (drought ) tolerance.
