Article S-Alkylation of Thiourea by Alcohols

You may use this procedure reported for methylation using methanol to form the salt in 70% yield

Heat a solution of thiourea (7.6 g) and HCl (4 mL) in methanol (30 mL) under reflux for 24 hours.

Condense the solution to a volume of 5 mL.

Cool the reaction mixture to 0 °C to give a white crystalline precipitate (1 g).

Collect the precipitate by filtration.

Wash the precipitate with cold methanol (2 ml).

Heat a mixture of thiourea (10.0 g, 0.13 mol), iodomethane (8.2 mL, 0.13 mol) and MeOH (100 mL) at 65 °C for 90 minutes.

Remove the MeOH in vacuum.

Transfer the resulting yellow solid to a sintered glass funnel.

Wash the resulting yellow solid with Et2O (5 x 50 mL) to obtain 2-methylisothiouronium iodide.
