The aim for my study is to prepare solid lipid nanoparticles stabilized with lecithin sodium deoxycholate and tween 80. The problem is that after the sonication process, and during the recrystallization I notice that the 2 phases (hydrophilic and lipophilic) were separated.
Does anyone have a solution?
The main problem that might be arising is due to Thermodynamic instability. When you decrease the particle size the other issue like Gibbs free energy also increases. If you are going for Probe sonication , I think its better to go for a cold water jacket, and you too need to think on the concentration of your surfactants as they play an important role to bring down your Free Energy. And also can keep slow mixing RPM so that the particles remains dispersed.
Dear Hassan, first i used ultra turrax at 10000 rpm for homogenization after i used probe sonication for 5 minutes but i didn't use an ice bath cz the Tm for the lipid is 33 degrees. Is there any steps that should i add for this protocol or not? Thanks
Well can give a try ..... I used normal water jacket for probe sonication while prepating nanoemulsion
Sonication will automatically increase the tempreture use normal water jacket @ room tempreture
Try to develop a condition to maintain the dispersion at room temperature so that it the thermodynamics is unaltered.

as said stabilizer concentration maybe a problem. If it is too high micelles and liposomes may cause depletion flocculation. On the other hand there maybe a problem with Ostwald ripening, which maybe triggered due to solubilization in surfactant micelles.
Yes, temperature maybe also a problem. Tween will be more hydrophobic at higher temperature (distribution between lipid, interface and water will change).
You may use 5 times 1 minute instead of 5 minutes with cooling in between.
I use the lecithin/deoxycholate/tween80 (55/25/20, w/w/w), i wil try the probe sonication with normal water jacket to see the effect on the stability of the system
