I am looking for a working protocol to modify the surface of silver NP with carboxylic acid (because I want to use EDC/ NHS crosslinkers to link my NP to antibody). I have found an article related to MNP (gold) where they only mentioned this ----- " MNPs were incubated in ethanolic solution containing 10uM MUA and 10uM MU". The author did not mentioned how long the incubation period is or how much percentage of ethanolic solution is required. In some other articles, I have found PEGylation to add COOH. Yet, I could not find any detailed protocol. I would highly appreciate anyone who could help me with this. Thanks

We used lauric acid to modify the surface of our Ag NPs and we are intending to use EDC/ NHS chemistry for conjugation to the antibody.Dr. Steingrimur Stefansson Thank you as well for your suggestion. We used lauric acid since we had this chemical in our lab.
I am wondering to know if you can give me any hints on how to quantify/ measure the conjugated antibody-NP. I came to know about microBCA assay. I can also use florescent secondary antibody and do microscopy. But, I am wondering to know any validated methods to confirm my conjugation is successful.
