what is the type or source of the sample. Are you talkign about the level in an entire crab ? Yuo can make an extract of a soluble solution nds then extract it with ether. can run that as suggested in an HPLC-MS or MS alone and monitor for the E2 molecular ion. the other comon way is to use an RIA for E2
The common method for E2 is RIA with sample extraction if you would like to measure this hormone in tissues. Firstly you should make homogenization of your samples and next extraction. You can use ether or hexane for extraction. I was working with Orion kits for E2 RIA but last time I am using CisBIO kits for E2. The antibodies are sensitive so you can use this analysis for low concentration of E2. Best regards, Hanna
