during spectrophotometer analyzing for a solution contains nanoparticles and protein I get 2 peaks at 420 and 280 nm . How can interpret these peaks ?

280 nm is band corresponds to the pi-pi* transition electrons in benzene ring of protein.
420 nm is plasmon bands of Ag nanoparticles. See our articles.
More specifically the 280 nm absorption is due to tryptophan and tyrosine.

Silver nanoparticles (AgNPs) gives an absorption peak in the range 400 nm to 480 nmٍ.Peak around 420nm belongs to AgNPs. Formation of nanoclusters or precursor you used may be caused a peak at 280 nm.

In my work, I get an absorbance peak at 420 nm for silver NPs on the UV-Vis scattering graph. Can I calculate their size directly without using electron microscopy or XRD?
