I am trying to differentiate neurons form neural stem cells isolated from SVZ zone of adult mice and I have two main problems:
Do you have experience with adult neural stem cells? What could be the critical point during the neurodifferentiation?

Neuronal cells tend not to grow much in culture, even if you start from progenitor cells. Strategies to increase yield include expanding them in the neurosphere state (avoiding terminal differentiation), using different substrates (Matrigel, PO, PLL, PA6 cells) and playing around with confluence. Higher confluence levels mean less divisions and, usually, less cell death.
There are several protocols and commercial neuronal media which you could try (stemcells, gibco, etc).
