Hi all,

I am trying to differentiate neurons form neural stem cells isolated from SVZ zone of adult mice and I have two main problems:

  • I culture NSCs as neurosphere but after 7-10 days the yield is very low. I am using knock out medium from gibco with glutamax and gibco neural supplement.
  • At the 2nd passage I seed 40x10^3 NSCs for cm2 in stem medium until the confluence. After that, I start the neurodifferentiation (Neurobasal medium supplemented with B27 and glutamax). After 3 days cells look pretty good, they start to take neuron-like morphology but apoptotic bodies appear. Apoptosis increases during the culture with massive cell loss around day 7.
  • Do you have experience with adult neural stem cells? What could be the critical point during the neurodifferentiation?
