How to identify the bacteria by morphologically
I have isolated some bacteria and their 16S RNA sequence showed that they belong to bacillus species and I want to perform some morphological tests for their identification. So which tests I need to performed and how?. If some share names of tests needed to perform and send me some manual for the practical performance. I’ll be grateful to him. Thanks
I am not an specialist in biology, but if the proble m is on image processing I could help you a litte. Please, send or show me some images of the types of bacteria that you want to analyze.

I don´t really understand what you neet, but here it goes:
The traditional tests for identifying bacteria based on mycroscopy and morphologyare:
- Dark field microscopy would identify spirils like Borrelia sp. Treponema sp., etc.
- Gram is routinely used for most bacteria: based upon color (red or purple) you can rave Gram-negative and Gram-positive (Color depend on membrane lipid constitution and permeability); based on forms you can seevariations like: cocci (Staphylococcus, Streptococcus etc) in diverse arrangements, like pairs, "grape", long strips etc; bacillus or rods, cocobacili, sporulated or not, intracelular or extracellular bacteria, etc.
- Ziehl Neelsen or Kynoiun - These are the standards for "Acid-fast" Bacili like Mycobacterium leprae and tuberculosis. It will stain most Mycobacteria and some special organisms. you will see them grouped and in pink color over a blue background.
They require nothing but dome specific stains, and an immersion field enabled microscope, plus some easy to obtain equipment. Therefore, if you already have some idea of the bacteria you´re dealing with, you can select your method.
Be aware that you cannot positively identify bacteria this way; at best you get presumtive identification, and you got to proceed with special microbiologic tests.
