I have done sequential extraction of leaf using petroleum ether, chloroform and methanol. how can i isolate compounds from my methanol extract. what type of compounds may present in it
You might have done preliminary phytochemical test. Based on result obtained you can see what kind of compound present in that plant. None can say or predict what compound may present without experiment but based on methanol polarity, semi polar compounds may be extracted. Moreover, depends on % meoh used for extraction.
For isolation of compound, there are many methods available but manual and simple step before subjected for HiFi technique, i suggest you to do TLC profiling, this will help you:
1. to detect compound presents
2. develop appropriate solvent systems for HPLC, MassSpectroscopy, NMR.
Dear Rajesh, the first thing you have to do to start a research is to make an intensive bibliographic research about the plant you are working, doing that you have an idea about the natural product you may have. If you don't find your specie you have to go into the genus. If unfortunately even in genus you don't find anything you have to do some color reaction to detect some kind of Phytochemical I.e. to detect if you have terpenes, flavonoids, alkaloids, and so on. By other side, the natural products will be extracted in many different solvents according its polarity. As all of my previous researchers have already provided ample amount of resource data & answered this topic in a perspective manner, initially I thought there is little left that can be further added to this topic. Then I thought it can be little heavy for one to go through all those data points to make his working plans more proficiently. So let me try to summarize this topic.
To start with I think Madam Olga has a very good point. Her point to go for review study of the selected drug is very primordial that works not only for this topic but almost for every research work. Through this review study researchers can find group of chemical compounds & their related activities [ if explored drug] that have been reported from a particular plant species.
Now to identify a group of compounds you can go for preliminary phyto-chemical screening to know whether your drug contains alkaloids, phenols, glycosides, terpenes or other group of compounds. For this purpose you can follow the reference book of Harborne as provided by Dr. Rao.
Chromatographic technics like TLC & HPTLC are also useful for further idendification of compounds.
FTIR is good for functional group recognization like OH, NH2, COOH, CO etc.
HPLC is a good tool for both isolation & identification of compounds
For simple isolation you can go for column chromatography with iso-cratic or eucratic solvent system or with preparative TLC method.
Now for structure elucidation of your isolated compounds you can go for NMR, LC-MS, GC-MS etc.
It depends on your interest, if it is an activity guided study the you need to check the active of the different components you have partitioned and work with the partition that gave the highest activity in which you need to do the TLC first to know the solvents that will give you the best separation of the different compounds in the partition then you can proceed to column chromatography and the process still continues....TLC... purification.
