I prepared a 5X of M9 minimal salt and 1000X elemental trace and mixed these 2 solutions to produce carbon-free minimal salt media. Biphenyl was used as a carbon source but was prepped separately with polar aprotic solvents such as acetonitrile, acetone, and DMSO. The concentration of biphenyl was 100 mg/mL.

The CFMM was mixed with the following volumes and reagents, and this would give me approximately 0.5% of Biphenyl in CFMM.

  • 20 mL of 5X M9 minimal salt
  • 10 μl of 100X or 1000X elemental trace
  • 500 μl of 100 mg/mL Biphenyl
  • 79.4 mL of autoclaved Milli-Q water.

The problem that I've encountered when I added 100 mg/mL of biphenyl (dissolved acetonitrile/acetone/DMSO) into the carbon-free minimal salt media, it (biphenyl) flocs and does not get homogenized with the CFMM (Carbon-free minimal salt media).

I've tried using milli-Q water as a solvent and heat the biphenyl to its melting point of 69.2 Celcius, and the result does not change when I added it to the CFMM.

What can I do to helps Biphenyl dissolute and homogenize with CFMM?
