I don't use Origin, but there must be a function for numerical integration to answer your question. For Area 1 integrate under the curve from 0 to Vb and subtract that result from the area of the rectangle containing Area 1. Similar for Area 2.
In your message to me you said the x-axis was days, but the plot shows a volume. Also, to be clear, you are asking for the areas of the two shaded regions - correct?
