I would like to calculate the amount of volatile analytes. I measured whit HS-SPME-GC-MS, but now I would like to use HS-SPME-GC-qTOFMS. Is it possible, that I cannot see my analytes on the chromatogram because of the LOD of qTOFMS?
I think it wouldd help if you told the analytes and the matrix. Of course everything is possible with som analyte... some will for example have so low vapour pressure that head space analysis (I guess that is what your HS stand for) does not work at all. additionanlly it might be the analytes have so little afinity to the SPME that this does not work.... etc etc..
I have already checked on the literature, that PDMS coating fiber should be good for microbial volatile organic compounds. I increased the extraction time and finally I found some compounds but still less as I expected.
To calculate the amount of volatile analytes in your samples you need a calibration curve for each compound. How to do it depends on the matrix. If you just noted that some compounds you sow with a single quad are not detected by you ToF system, consider that ToF-MS sensitivity is 1 or 2 order of magnitude lower (or even less) than a single quad MS.
