First, you need to perform a calibration curve of Ascorbic acid. I normally use 100,200,400,600,1000 μM solutions of Ascorbic acid.
Second, perform the same measurements with your extract (dilution may be needed) in a concentration that fall inside the range of the calibration curve (some trial and error may be necessary).
Finally, perform linear interpolation by using the calibration curve of the Ascorbic acid and the absorbance value of you experiment with the extract.
the results are usually presented as μmol equivalents of Ascorbic acid per gram of extract.
I was just goingto ask the same question but this time it concern with testing of guava and the amount ascorbic acid equivalents to test for calibration curve or standard curve.
