We are interested in purchasing the Leica CryoJane tape transfer system to facilitate sectioning of murine bone (primarily tibia and femur). However, until we have funding for this equipment we are trying to create our own "DIY" version.
We can successful cut sections which adhere to the tape. The problem is that transfer of the sections from the tape to the slide is inconsistent - sometimes giving beautiful sections and on other occasions partly or not sticking.
We are trying to figure out whether the UV (source / duration) or the slide temperature (as the CryoJane has a temperature controlled pad) are key for improved reproducibility.
If anyone has managed to do this or can advise we would be most grateful.

You could use Japanese tape instead http://section-lab.jp/ We use this for undecalcified bone all the time (see any of my recent pubs or those from David W Rowe's group). The sections stay on the tape and you paste the tape onto a slide using an adhesive. I think Section lab only sells large quantities of tape at a time, but our molecular core here in CT does sell to outside people, if you can figure out a way to pay them! It's quite expensive, but will work with your existing equipment, and its pretty easy to get nice sections. I can send you a contact if you are interested.
I don't know anything about the intensity of the UV for the cryojane, but the slide is very close to the light and it just does a short flash. From memory, it's easier to get nice transfer with the 4x slides compared to the 1x. If you are doing brightfield imaging the adhesive on the slide looks bad, so it will be difficult to get nice images if you are interested in the periosteum.

This is actually not an aswer but a question to Nicholas and all the others that can help though.
We are in the same situation, not having enough now to purchase the whole thing, but I think 4x slides and the tape windows are more than affordable!
Do you simply cut the samples onto the tape, place them on the slide (which T° do you keep them?) and UV light? for how long and using which source?
I've gone through the sites there but I think it's easier just to buy at least the two things I mentioned from Leica directly..

What you describe has partially worked in our hands - it may depend on the type of tissue that you wish to section. Even with the actual CryoJane we experience a small amount of consistent bone marrow loss from sections of murine long bones. Due to this we are examining other options and would suggest that you also look at Brya's response above . thanks for the prompt answer! I have checked Brya suggestion, but I thought it could have been easier just to ask Leica instead of having stuff shipped from Japan xD
In general, we focus more on femoral condyles and tibia, so overall the knee joints.so I reckon the BM loss is not that important to us! Anyway my mail is mario.alvarez dot qmul so you can drop me an email and we can further discuss this!
