When you would like to observe TEM of biological samples, better to do at lower voltage or in cryo-mode...so sample preparation would be critical to observe the specimen...apart from that as the scale is in micron, the rod type structures are also large, not in nano regime for sure...i suggest you do some high resolution SEM first then go for TEM followed by diffraction of rods...
Reham Hamida In TEM there are different mode of operation, one is imaging, another is electron diffraction, and the third either elemental analysis like EELS or texture analysis... so my point is when you do TEM imaging, once you see rod shaped crystalline structures, you could take diffraction on that rod itself to know the composition or type of crystal and its orientations...100X or 100000X? later one you should see detail texture of rod or any shape...
