Hello. I have to isolate, from lake water, flavobacteria and/or bacteria with the gene bla CTX-M for my theses. I want you advise on this purpose, if you have a protocole, a way to do that .
I would take a large volume of Lake water (depends on the water quality), filter it using a bacterial filter and then place it on a plate with and without Ampicillin. Than you simply pick one colony which is growing on Amp and streak it on a fresh Amp plate. You could screen for bla CTX-M by PCR

I had the same idea as yours. Now I have to choose the appropriate culture medium.
The lake is in North Italy : the lake Maggiore near to Milano. It isn't realy contaminated in heavy metal but there is many antibiotics resistances from bacteria community, especially that of Flavobacterium community (which I did not a lot of knowledge).
For isolation of CTX-M positive E. coli, process the water as described above and spread 10ul of this on EMB agar containing 1ug/ml cefotaxime. And then confirm using a gene specific PCR
