I would like to quantify the nascent RNA transcribed in my mouse model after EU admin. and chase. I would like to use the Click-iT Nascent RNA Capture kit from Thermo to do this, but am wondering how others have successfully modified the protocol for use in tissue samples? The protocol that comes with the kit is written only for in vitro application, and I only know of one paper that uses this kit for in vivo expression, and their protocol is not very descriptive in the paper they published. The protocol that comes with the kit is also a bit confusing in my opinion.
