We're trying to use eppendorf femtojet microinjector for pronuclear manipulation in mouse embryo. The trouble is, after loading the mix in the needle, we are always ending up with air bubble which is blocking the manipulation.
Is there any trick to avoid getting air bubble or remove the bubble?

Hi Kashfia,
In case the mix is viscous or sticky you may need to load the mix from the back of the injection pipette using a fine needle (30G) then you push (using pressure from femtojet) or any other micromanipulator plunger.
If the mix is light or less not viscous, you can aspirate from the tip of the pipette. But you should make drop of the mix in the injection dish.

Thank you Mulyoto. Someone also told me that it also matters based on the volume of the mix (1-2uL vs. 10uL) - do you have any insights on that?
