I would like to try this technique in wheat breeding.
Does it work well ? Do you cut the top of the glumes, lemmas and paleas before diving the spikes in water ? What is the perfect duration and temperature of this handling?

Though I have never tried this technique but the success of it depends upon differential senestivity of anthers and stigma to hot water, which I think is not there

It is somewhat feasible but only for stage specific tillers (inflorescence) and one can go for it if nothing else is working. Female part also gets adversely affected.
I never did that but the extraction technique is very practical. Perhaps in large quantities to water technique is required!

In fact, I had found this technique in the article from Makhdoom Hussain and al. and tried it in the corner of the lab on a very little sample of wheat tillers from a same genotype but it gave very different responses depending of the spikes and their stage. But as I said, it was a very small sample and i think i will try it later more seriously on a larger scale.

Anice, this method (hot water treatment) has been applied by Japanese rice breeders for long time. At least since 1940s.
