I stained collagen rat tail I with NHS Fluorescein . I tried looking at the stained fibers using normal fluorescence microscopy.The fluorescence image looked blur and did not see any fiber. Has anyone worked on this ?
I have not worked on confocal yet. I have only tried to see the fibers using normal fluorescence microscopy. Do you think we can see the fibers using this technique?
I am not familiar with fillers. But I tried to stain the collagen solution and after gel formation , exposed to 494nm wavelength. I can see the green fluorescence image (mostly blur green) but do not see any traces of fiber or any structure that looks like fiber. May be I need to use thin layer of collagen gel at my 96 well plate.
Also, I did not find any paper on collagen fiber using epi fluorescence microscopy.
You can check out second-harmonic generation microscopy. You should also be able to see some of the collagen structure with normal confocal under reflective mode.
