Earthworms were stored at minus 80 degreed freezer. I need to extract the gut from earthworms for microbial analysis. However, i am having problem to extract gut because the earthworm became wet from inside after defrosting and its really hard to pull out the clear guts.
For understand the gut microflora you might want to use fresh earthworms.
After their you sill them in ethyl alcohol . Surface disinfect them with 10% blecah for 4 minutes.
Rinse them with 1 min sterile distilled water.
Take 1 cm section which is than aseptice macerated and plated on steril media.
This will be done on the length of the warm noting the sequence.
The organisms identified should be from the gut microflora I believe.
Hello Kaushik; Paul's method avoids the problem you encountered. Since the worms' gut is suspended by the septa, you would have a problem getting it out intact. Best regards, Jim Des Lauriers
