I am studying GPR39 in differentiated neuroblastoma cells. GPR39 has a molecular weight of 51kDa, I am getting a week band on 51 kDa but a stronger band on 70 kDa. Any ideas on what happened?
To interpret results from any Western blot experiment, a loading control and positive control are critical.
A loading control accounts for potential variation in:
- the amount of protein sample loaded in each lane,
- protein transfer efficiencies from the gel to the membrane among different samples/lanes,
- antibody incubation (for primary or secondary antibody), and signal detection across separate samples/lanes.
The signals from loading controls are typically used to normalize the signals from the proteins of interest. To use a loading control for these purposes detection with control protein antibody and the experimental antibody should be done on the same blot.
Beta actin is ideal loading control.
Positive control is critical to validate the antibody.
I recommend you to repeat the blot, runong beta actin and use positive control.
