Fungal Endophyte gDNA Quality: Old vs. New Leaves, and Large vs. Small Leaves?

I am extracting endophytic fungal gDNA from the leaves of several Myrtaceaous species.

How may leaf maturity and age affect the endophytic fungal gDNA quality as well as fungal diversity? Would the similar differences exist among leaves of different masses within the same plant taxa?

It seems hard to answer to your question, but befor leaf gDNA extraction, I sugget you to isolate culturable endophytic fungi on different culture media like MYP and .., it gives you more clear answer about leaf age differences then you could decide which age is suitable and then by leaf gDNA extraction sequences, you also could campare total population with previus results of culturable endophytes.
