I am facing issue trying to prepare water in oil emulsion via microfluidic system using 190um quartz-made chip.

The continuous phase is made of 8% polymer / DCM / 1%Span80 and the droplet phase is made of 1% hydrophilic API in water.

It seems that my organic phase is not able to break the aqueous flow to generate droplets. The organic phase accumulates as droplet in the outlet channel (after the junction) and the water phase remains in the middle (See attached screenshot). It ends in an unstable O/W emulsion instead of W/O emulsion.

I thought the issue was coming from SPAN80 forming a film in my outlet channel so I tried to wash it several times with DCM and EtOH, but same issue remains.

Besides, I also tried to exchange phases to prepare an O/W emulsion, but same observations.

Any advice would be highly appreciated !
