I had a question about DRUG-seq as described in Ye et al. 2018 (https://www.nature.com/articles/s41467-018-06500-x)"

The protocol is described as:

1. Lysis and RT to add barcodes at the 3' end of the cDNA

2. Pooling and preamp

3. Tagmentation to add Illumina sequences.

My question is: Doesn't tagmentation remove the barcodes from the cDNA? I thought tagmentation will add new ends to the cDNA, removing the barcodes.

What am I missing?
