It should. I have tried 10 mM NaCl, 10 mM HEPES on both sides of the membrane, for example, in patch clamp. The effect may depend on the particular channel however. Also, in your case you seem to have low osmolality in conjunction with low ionic strength.

Here is a publication dealing with ionic strength, also using patch clamp: Pflugers Arch. 2000 Jan;439(3):315-20. Reduction of ionic strength activates single volume-regulated anion channels (VRAC) in endothelial cells.

Regarding the citation above: It should be noted that VRAC (volume-regulated anion channels) are a special case, as they are activated specifically by low ionic strength, as well as cell volume changes (hence the name). See: Pedersen, S.F., Okada, Y. & Nilius, B. Pflugers Arch - Eur J Physiol (2016) 468: 371. So VRAC (if present) would certainly be activated in the above conditions, but probably aren't a good indicator of ion channel activity in general.

I agree though, in principle there should be nothing stopping a ligand-gated channel from opening under those conditions. Although, since your concentrations of KCl are equal on both sides, you'll of course need to apply a holding potential to see any currents. Also, ion mobility can differ somewhat from normal in extremely dilute or extremely concentrated solutions, which could present a complication.

@J. Ashot Kozak: The ionic strength you used is 2-order higher than one I used. Do you still think that I can see ion channel activity of serotonin receptor under the conditions? And low osmolarity might affect ion channel activity?

@Thomas Murphy: I am using confocal fluorescence microscopy to check ion channel activity (Cacium indicator + Cacium ions diffused in GUV) without holding an electric potential. Could you recommend me the ways to increase the diffusion rate of calcium ions through ion channels?
