No, simply because they are sporeformer, spores could survive for years, the evident and good example for this the sample that you have collected from volcanic soil. my advise always keep a stock culture (pure) of your isolates, in this case you guarantee no contamination, in the same time you can use this when ever you want and they will grow in shorter time compared to isolating process.

I fully agree with the statement made by Dr. Albaser. Actinomycetes (actinobacteria) can tolerate high temperature and they have the capacity of more adaptability than bacteria and fungi. Moreover, almost all are spore former therefore there is meager chance to reduce there population during soil storage period for a longer time.

Google it on google image, if you trying to compare it then you should carry biochemical and genetic (DNA) characterisation.
