Dear all,
The technical bulletin for the Iron Assay Kit SIGMA MAK025 mentions that this kit allows the quantification of total intracellular iron.
However, it is not clear whether this includes haem iron?
In the litterature, authors state they quantify total intracellular non-haem iron or alternatively total intracellular iron, using this kit....
May somebody help?

The catalogue and information is not available in my Country as usual , but as far as i guess / Know: Lipoxygenases (LOX) are non-heme iron-containing dioxygenases.
Parisa Ziarati Thanks a lot for your answer! However, you are likely talking about the Kit SIGMA MAK363 (not MAK025). I will try to add the file corresponding to the MAK025 kit.

Dear Parisa Ziarati , we have asked SIGMA , and they did not appear very convinced in their answer that heme- iron should NOT be quantified using this kit. In addition, in our hand, we suspect that heme-iron was indeed quantified (comparing myoglobin deficient tissus versus control tissus).
Conversely, the authors of a recent Journal Pre-proof "SLC46A1 contributes to hepatic iron metabolism by importing heme in hepatocytes" DOI: https://doi.org/10.1016/j.metabol.2020.154306 state that:
"Cellular iron content (non-heme iron) was determined by an Iron Assay Kit (Cat. MAK025, Sigma-Aldrich)"

But the not transparency of the Sigma -Aldrich .....is not good. Anyway, sorry to not be helpful in this case.
