I am working on grafting of Folic acid on polymeric nanoparticles exploiting the NHS/EDAC chemistry. The polymeric NP is functionalised to provide primary amine for the amide formation. During the reaction between the NHS-activated Folic acid and functionalised NPs, the amide bind is formed. What could be the ideal temperature for this step reaction? Providing temperature play any key role in the conjugation and amide bond formation ? Or room temperature is enough (25-35°C) for this reaction.
