I'm using DTT for reduction of disulpide bond. At the same time I'm running the reaction under N2 gas. My question is wether there are any effects of purging N2 on the total activity of DTT as reducing agant for disulphide bond ?
However, DTT will loose its reducing capacity relatively quick, so for example if you are using a stock solution of DTT that was prepared "a while ago", you may not have sufficient reducing capacity.
Also, DTT in solution will oxidize relatively quickly even when frozen, so it is a good idea to always prepare fresh.
