Hi everyone! I wonder if anyone could indicate me an efficient and simple protocol for separation of leaf tissues (epidermis, mesophyll, vascular tissue) that could be used for crops, such as tomato, soybean, maize, etc.?
I appreciate any suggestion given.

Do you mean machine vision segmentation or physical separation from samples?
Protocol for leaf isolations.
Use a surface disinfection 10% chlorox bleach in aqueous distilled water.
Time of 4 minutes with a 1 minute rinse in sterile distilled water.
Take 10 leaves stacked and core them with 1 cm diameter cork borer.
The 200 cores are surface disinfected and used for plating on potato dextrose agar.
Borer circles are dried under the laminar flow sterile hood. The circles are plated in the Potato dextrose medium 5 per 9 cm petri plate. The plates per replication and 4 replicates to give the appropriate sample size and ability to calculate experimental variation.
