You can dilute your bacteria in liquid culture (or water) as 1:100, 1:1000...1:10^6

Then, you can finally spread 100 uL of final diluted tubes onto the agar media.

Then, next day, you can coun how many bacteria you have.

You can multiply 10^6 * you bacteria to calculate your CFU/mL.

I agree with Karen A. Darbinyan but the 9 ml of broth. The mean (broth) is peptone water 1 gm / 1 L water or saline solution 0.085 % of NaCI.
