I am currently working on Pluronics as non-viral vectors for gene delivery. I am combining Pluronis with a cationic polymer, and would like to know the proper ratio for this complex to get an effective nanoparticles. Do I make the ratio to the pDNA or to the cationic polymer?

this is a very intersting question. We also tried to encapsulate DNA with PEI-PCL-PEG triblock copolymers with various ratios. We try to adjust the positive charges of PEI with the negative charges of DNA. Although we used an high excess of positive charges - we still saw free DNA in the gel.
