I want to measure the total carotenoid content of watercress leaves. I'm using 25μg powder of freeze-dried watercress and 10ml of acetone for the extraction process. I'm measuring the absorbance in a UV-Vis Spectrometer. Which are the best equations to use for the correct quantification of total carotenoids?What about the Wellburn and Lichtenthaler ones? Information in the literature is a mess. It seems that everybody is using whatever they think is correct.
