I want to adoptive transfer some cells into recipients' thymus. But I have no experience on that. Can anyone who has some experience on this method tell me how to do this experiment?
What animals are you working with? We did intrathymic injection with FITC on mice over 10 years ago. You should be able to adjust the protocol to transfer your cells (in stead of FITC in our case) into the thymus in a similar way.
First you should inject the anesthesia (check your institute what can be used) to leave the animals sleep. Put them on the dissecting stage and open the chest after cleaning the skin, and expose the thymus. Inject the cell suspension slowly. After done, close the chest with surgical clip. It seems very brutal, but the animals recovered very well in our case. If you are working with larger animals, you might need to use anesthesia machine with breathing help.
Since it was 10 years old, I am not sure if new technique has evolved (not working in this field for a while). Hope this helps. Good luck.
I work with B6 mice. When you talk about exposing the thymus and injecting the cell suspension slowly, you mean directly Insert the needle into the thymus and inject the cell suspension?
