Using RSM, What is the best for fermentation conditions/medium optimization; make a broad screening of factors by PB design then optimize by Central Composite Design or make preliminary studies for each factor alone using one factor-at-a-time then choose the critical factors and optimize them by Central Composite Design??
You always want to use some type of Designed Experiment over OFAT. The type of screening design you need is the question. A PB design generally uses 2 levels per factor. If you have more than 2 levels per factor, you may want to use an optimal factorial design or a split plot design.
OFAT -> PBD -> BBD/CCD (RSM) -> Genetic algorithm is the preferred method for media optimization.
It is better to go with the one factor at a time optimization, then PB/Taguchi and finally RSM, but if you are confident enough, you can directly go for RSM after OFAT.
OFAT is a waste of time. A PB or OFD should be the first step. OFAT uses too many samples and does not give you a good idea about what really is significant nor the interactions between factors. A 20 run, 10 factor PB design is far more powerful than the corresponding t-tests and gives the researcher up to 19 pieces of data. OFAT has the distinct disadvantage of not finding small differences and not being very powerful.
Follow Simplex or RSM to arrive at the optimal condition with minimum experiments. Design Expert Software is useful for the analysis.
Selection of technique or tools will depend upon the media composition one is going to use. If, the media contains more than 5 component it is better to go for PBD, otherwise, one can start using classical one point at a time technique. Both the technique have their own drawback. However, i would suggest to go with classical technique if one is handling five or less no. components in media. Otherwise, go for PBD and then confirm using classical technique prior to RSM.
