I am trying to separate suspension of PHB particles, stabilized with PVA. The DLS results show two populations with peak maxima about 150 nm and 380 nm. The Zeta potential was -3 mV (pH 7) across the range from 0 to 150 mM of NaNO3. For separation, I am using Asymmetrical flow Field flow fractionation with SC (250W) separation channel and PES and RC membrane (10 kDa). I tried different mobile phase (0; 25; 50 and 150 mM NaNO3), addition of PVA (0.005 hm%) or SDS. Probably, the particles are interacting with the membrane. On the Flow-through (cross flow rate 0 ml/min) I am getting some small signal from LS detector. But for the cross flow rate 0.1 ml/min the particles are retained in the channel and do not elute at all, not even during rising part. Any ideas for steric-stabilized particles/mobile phase/conditions?
