I want to study naive CD4 T cells polarization and therefore I need to be sure that the cells I will culture will be naive. So far I have found isolating kits from Miltenyi, STEMCELLS Technologies and R&D. Does anyone else use a different kit?
Do you remember the percentage of naive cells purity? Would you recommend the Miltenyi kit? I think about that option because I dont want to torture my cells through FACS.
I have used the CD4+ CD62L+ kit from Miltenyi to isolate naive CD4 from mouse spleen and it works well. I would look to see the number of CD62Llo cells in your lymph node prep to see if you could just isolate CD4+ T cells without having to use the more expensive CD4+CD62L+ kit.
Yes I see. I would sleep quietly if that kit used a anti-CD44 for the isolation, but still (based on the datasheet) I think it would be reliable. I will check what you said. Thanks!
@Jonas Cicenas. Yes I did but we would need to buy the ecquipment as well and we can't afford it at the moment. So, I'm looking for cost-effective options.
