Actually, Cy7 NHS ester can be used in staining peptides. I want to stain the peptides that can bind to monocytes. Hence, I used Cy7 NHS ester for labeling the peptides and DAPI for labeling monocytes. However, I found that monocytes have red fluorescence which symbols Cy7 NHS ester. Is it possible that monocytes can bind Cy7 NHS ester?
Is this a common method cause I'm not sure how this would technically work? The Cy7 NHS ester reacts with amines so it will label proteins on the surface and in the cell.
Amine-reactive dyes are even used as viability markers since live cells will have dim staining (surface) while dead cells will show bright staining (entire cell). I think it's more common to try and tag your epitope of interest (though that could mess it up) and you have to know the peptide sequence beforehand.
